Anticonvulsive and Antioxidant Effects of Pioglitazone on Pilocarpine-induced Seizures in Mice

Background: Epilepsy is a neurological disorder caused by uncontrollable discharge of action potentials from neurons in the brain. After seizure, oxidative stress may cause significant neuronal damage. In current study, we assessed anticonvulsant and antioxidant properties pioglitazone, peroxisome proliferated activated receptor-γ (PPAR-γ) agonist that used type-2 diabetes, on pilocarpine-induced seizure mice. Methods: Pilocarpine (400 mg/kg) or normal saline was injected intraperitoneally 4 hours after oral administration Pioglitazone (80 mg/kg). Also, carboxymethyl cellulose administered orally control groups. all mice were observed for 1 hour to measure latency time. Pilocarpine-induced seizures categorized using Racine scale. Then animals decapitated, brain removed hippocampus dissected. Finally, level Malondialdehyde (MDA) Catalase (CAT) activity, Superoxide Dismutase (SOD), Glutathione Reductase (GR) levels quantified biochemical methods. Results: significantly increased onset stages 1-4 (P≤0.01-0.001). pioglitazone prevented development stage 5 seizure. decreased MDA (P<0.01) elevated CAT (P<0.01), SOD GR (P<0.001) enzymes compared those pilocarpine group. Conclusion: The findings this study indicate effect play an important role its protective effects against damage
